Cross-talk between Stat5b and estrogen receptor- and - in mammary epithelial cells

چکیده
Both 17 -estradiol and prolactin play important roles in the mammary gland, raising the possibility of functional cross-talk between the two signaling pathways. Here, we demonstrate that estrogen receptor(ER ) and (ER ) are both able to potentiate transcription from a Stat5-responsive promoter when activated by prolactin. Potentiation was observed not only in the presence of 17 estradiol, but also in the presence of anti-estrogens such as tamoxifen and ICI 182,780. The magnitude of the response was dependent on cell-type: in the HC11 mouse mammary epithelial cell line ER potentiates transcription efficiently whereas ER showed low activity. Conversely, in COS-7 cells, both estrogen receptors were active. We show that activation domains in the N-terminus (AF-1) and the C-terminus (AF-2) of the ERs are dispensable for potentiation. The effects are dependent on the presence of an intact DNA-binding/hinge domain, which we show is capable of interacting with Stat5b in vitro and in HC11 cell extracts. We conclude that ER and ER act as coactivators for Stat5b through a mechanism which is independent of AF-1 and AF-2. Journal of Molecular Endocrinology (2001) 27, 93–106

Estrogen-induced mammary fibroadenoma in a lamb: hormonal and immunohistochemical aspects
A fibroadenoma was diagnosed in the mammary gland of a lamb by immunohistochemical method andmeasurement of oestradiol hormone. The tumor was characterized by an encapsulated firm mass with acreamy-white cut surface. Histologically, it consisted of variably-sized sinus ducts, covered by a single ormultiple layers of proliferated epithelial cells, and embedded in a loose connective tissue. Immun...
متن کامل17β-Estradiol and ICI182,780 Differentially Regulate STAT5 Isoforms in Female Mammary Epithelium, With Distinct Outcomes
Prolactin (PRL) and estrogen cooperate in lobuloalveolar development of the mammary gland and jointly regulate gene expression in breast cancer cells in vitro. Canonical PRL signaling activates STAT5A/B, homologous proteins that have different target genes and functions. Although STAT5A/B are important for physiological mammary function and tumor pathophysiology, little is known about regulatio...
متن کاملبررسی توزیع گیرندههای استروژن و پروژسترون در ضایعات Epulis Fissuratum
Statement of Problem: Epulides Fissurata (EF) are common proliferative and denture- induced lesions of the oral cavity with a predilection for female. This suggests a possible role for sex steroid hormones in the development and progression of these lesions. Purpose: The objective of this study was the immunohistochemical evaluation of epulis fissuratum of the oral cavity for estrogen and proge...
متن کاملCross-Talk in the Female Rat Mammary Gland: Influence of Aryl Hydrocarbon Receptor on Estrogen Receptor Signaling
BACKGROUND Cross-talk between the aryl hydrocarbon receptor (AHR) and the estrogen receptor (ER) plays a major role in signaling processes in female reproductive organs. OBJECTIVES We investigated the influence of the AHR ligand 3-methylcholanthrene (3-MC) on ER-mediated signaling in mammary gland tissue of ovariectomized (ovx) rats. METHODS After 14 days of hormonal decline, ovx rats were ...
متن کاملBovine mammary stem cells: cell biology meets production agriculture.
Mammary stem cells (MaSC) provide for net growth, renewal and turnover of mammary epithelial cells, and are therefore potential targets for strategies to increase production efficiency. Appropriate regulation of MaSC can potentially benefit milk yield, persistency, dry period management and tissue repair. Accordingly, we and others have attempted to characterize and alter the function of bovine...
